==== Emulsions ==== The so-called "oil" adjuvants are really oils intended to be made into an emulsion with water, either oil-in-water (O/W) or water-in-oil (W/O). They would not work when moved out of the droplet form. Freund's complete adjuvant is a solution of inactivated Mycobacterium tuberculosis in mineral oil and/or lanolin developed in 1930. It is used as water-in-oil, It is not safe enough for human use. A safer version without the bacteria is known as Freund's incomplete adjuvant, but its effect is weaker. In either case, the W/O structure probably helps vaccines release antigens for a longer time. Despite the side effects, its potential benefit has led to a few clinical trials, including a few attempts at adding other microbial-derived parts to balance safety and strength. (A common side effect for both versions is long-term retention of the oil in tissue.) All "oil" adjuvants approved for humans are oil-in-water. They adsorb antigens to their surface and induce immune cell recruitment. All of these human-approved O/W adjuvants use squalene, an oil produced by human liver and skin. MF59 is an oil-in-water emulsion of squalene adjuvant used in some human vaccines. As of 2021, over 22 million doses of one vaccine with squalene, FLUAD, have been administered with no severe adverse effects reported. In addition, squalene-based O/W emulsions have also been shown to stably incorporate small molecule TLR7/8 adjuvants (e.g. PVP-037) and lead to enhanced adjuvanticity via synergism. ==== PC reception ==== Upon its retail release, Counter-Strike received highly favorable reviews. The New York Times reported that E-Sports Entertainment ESEA League started the first professional fantasy e-sports league in 2004 with the game Counter-Strike. Some credit the move into professional competitive team play with prizes as a major factor in Counter-Strike's longevity and success. The game was praised by critics for its emphasis on teamwork and communication among players. Scott Osborne, writing for GameSpot, felt that the audio messages were a decent feature, which covered requests for backup, status reports and warnings. One problem, according to him, was that both teams used the same voice. Clayton Wolfe of IGN expressed the importance of teamwork and stating, "It's the team-based tactics that make [Counter-Strike] so fun." Computer Games Magazine's Bill Hiles also commented about the social aspects of the game, saying that a player should work in teams rather than alone. Joe Dodson for GameRevolution wrote that "Nothing can beat a coordinated, experienced team in [Counter-Strike], except an even more coordinated and experienced team." He also found the team talk favorable. Most reviewers agreed that the GoldSrc engine felt dated, but still praised the graphics. Hiles commented that the visuals were acceptable and "do an admirable job portraying outdoor locations", while Dodson noted the graphics were identical to Half-Life.
=== Peptides with similar functions === In 2001, biologically active hydroxyproline-rich glycopeptides were isolated from tobacco which activated the production of protease inhibitors in a similar way to systemin in tomatoes. Although they are structurally unrelated to systemins, their similar function resulted in them being named hydroxyproline-rich systemins (HypSys). Following the initial discovery other HypSys peptides were found in tomato, Petunia and black nightshade. In 2007, HypSys were found outside the Solanaceae, in sweet potato (Ipomoea batatas) and sequence analysis identified HypSys analogs in poplar (Populus trichocarpa) and coffee (Coffea canephora). Systemins are highly conserved between species, whereas HypSys are more divergent but all contain a conserved proline or hydroxyproline-rich central domain. In 2006, AtPEP1, a 23 amino acid polypeptide was isolated from Arabidopsis thaliana, which was found to activate components of the innate immune response. Unlike HypSys, AtPEP1 is not post-translationally modified by hydroxylation or glycosylation. Six paralogs of the precursor have been identified in A. thaliana as well as orthologs in grape, rice, maize, wheat, barley, canola, soybean, medicago and poplar, although the activity of these orthologs has not been tested in assays. The predicted structures of the paralogs of AtPEP1 are varied within A. thaliana but all contain a SSGR/KxGxxN sequence motif. For example, tubulin is formed by the dimerization of α-tubulin and β-tubulin and this dimer can then polymerize further to make microtubules. == Genetics == Spondyloperipheral dysplasia is one of a spectrum of skeletal disorders caused by mutations in the COL2A1 gene, located on chromosome 12q13.11-q13.2. The protein made by this gene forms type II collagen, a molecule found mostly in cartilage and in the clear gel that fills the vitreous humour (the eyeball). Type II collagen is essential for the normal development of bones and other connective tissues (the tissues that form the body's supportive framework). Mutations in the COL2A1 gene interfere with the assembly of type II collagen molecules. The protein made by the altered COL2A1 gene cannot be used to make type II collagen, resulting in a reduced amount of this type of collagen in the body. Instead of forming collagen molecules, the abnormal protein builds up in cartilage cells (chondrocytes). These changes disrupt the normal development of bones, leading to the signs and symptoms of spondyloperipheral dysplasia. The disorder is believed to be inherited in an autosomal dominant manner. This indicates that the defective gene responsible for the disorder is located on an autosome (chromosome 12 is an autosome), and only one copy of the defective gene is sufficient to cause the disorder, when inherited from a parent who has the disorder.
Piperacillin is a broad-spectrum β-lactam antibiotic of the ureidopenicillin class. The chemical structure of piperacillin and other ureidopenicillins incorporates a polar side chain that enhances penetration into Gram-negative bacteria and reduces susceptibility to cleavage by Gram-negative beta lactamase enzymes. These properties confer activity against the important hospital pathogen Pseudomonas aeruginosa. Thus piperacillin is sometimes referred to as an "anti-pseudomonal penicillin". When used alone, piperacillin lacks strong activity against the Gram-positive pathogens such as Staphylococcus aureus, as the beta-lactam ring is hydrolyzed by the bacteria's beta-lactamase. It was patented in 1974 and approved for medical use in 1981. Piperacillin is most commonly used in combination with the beta-lactamase inhibitor tazobactam (piperacillin/tazobactam), which enhances piperacillin's effectiveness by inhibiting many beta lactamases to which it is susceptible. Many other physiologic changes occur under the control of progesterone and estrogen. Adenosine monophosphate deaminase deficiency type 1 or AMPD1, is a human metabolic disorder in which the body consistently lacks the enzyme AMP deaminase, in sufficient quantities. This may result in exercise intolerance, muscle pain and muscle cramping. The disease was formerly known as myoadenylate deaminase deficiency (MADD). In virtually all cases, the deficiency has been caused by an SNP mutation, known as rs17602729 or C34T. While it was initially regarded as a recessive (or purely homozygous) disorder, some researchers have reported the existence of similarly deleterious effects from the heterozygous form of the SNP. In the homozygous form of the mutation, a single genetic base (character) has been changed from cytosine ("C") to thymine ("T") on both strands of Chromosome 1 – in other words, "C;C" has been replaced by "T;T". A rarer but analogous condition, in which two guanine bases ("G;G") bases (in the unmutated form) have been changed to adenine ("A;A") has also been identified. While there has been no consensus on the effects of the heterozygous form – either "C;T" or "A;G" – some evidence has been found that it too has caused AMPD1 deficiency. In addition, some sources have suggested the existence of a rare, acquired form of AMPD1 deficiency.
